摘要
目的观察七氟烷对慢性阻塞性肺疾病(COPD)大鼠肺组织中P65及肺功能的影响,探讨七氟烷的肺保护作用及相关分子作用机制。方法将32只清洁级Sprague-Dawley大鼠随机分为正常组、COPD模型组、1%七氟烷干预组及2.5%七氟烷干预组,每组8只。采用被动吸烟复合气道内注射内毒素法制作COPD模型。模型建立后将七氟烷干预大鼠投入密闭管路连接的动物麻醉诱导盒内,分别予1%及2.5%七氟烷持续吸入1 h,然后采用大鼠肺顺应性测定软件1.0分析各组肺功能参数,并进行组间比较。采用免疫组织化学法及电化学发光(ECL)-Western印迹法测定大鼠肺组织P65水平,运用Image-Proplus 6.0软件行半定量分析,比较各组肺功能及P65表达水平。结果 2.5%七氟烷干预组的气道阻力显著低于COPD模型组(P<0.05)。1%七氟烷干预组、2.5%七氟烷干预组的肺顺应性均显著高于COPD模型组(P值均<0.05)。1%七氟烷干预组、2.5%七氟烷干预组P65的表达均显著低于COPD模型组(P值均<0.05)。结论七氟烷能有效降低COPD大鼠气道阻力并改善其肺顺应性,具有较好的肺保护作用。七氟烷可能通过抑制P65表达发挥其肺保护作用。
Objective To observe the influence of sevoflurane on pulmonary P65 expression and lung function in rats with chronic obstructive pulmonary disease(COPD),and to discuss its pulmonary protective effects and the related molecular mechanisms.Methods Thirty-two male Sprague-Dawley rats was randomly divided into control group,COPD group,1% sevoflurane COPD group and 2.5% sevoflurane COPD group,with 8 rats in each group.COPD model was established by passive smoking and tracheal instillation of LPS.Rats in sevoflurane groups were put into anesthetic induction containers connected by closed tubes and inhaled 1% or 2.5% sevoflurane for 1 hour.All rats underwent lung function test by lung compliance analyzer 1.0.The expression of P65 was observed by immunohistochemistry and Western blotting analysis.Semi-quantity analysis was performed using Image-Pro-plus 6.0 software.The differences of lung function and P65 expression levels were analyzed and compared in all groups.Results The airway resistance in 2.5% sevoflurane COPD group was significantly lower than that of COPD model group(P〈0.05).The lung compliances in 1% and 2.5% sevoflurane COPD groups were significantly higher than that in COPD model group(all P〈0.05).Furthermore,the expressions of P65 in 1% and 2.5% sevoflurane COPD groups were significantly lower than those in the COPD model group(all P〈0.05);no significant differences were found between the former two groups(P〈0.05).Conclusion Sevoflurane can effectively decrease the airway resistance of COPD rats and improve their lung compliance.It can protect the pulmonary function through depressing the expression of P65.(Shanghai Med J,2010,33: 532-535)
