摘要
目的观察四物汤配方颗粒对骨髓抑制小鼠造血功能的影响,探讨其对抑制小鼠骨髓基质细胞内转化生长因子β1(TGF-β1)和细胞信号内转导分子果蝇MAD类似基因(Smads)表达影响。方法将50只BALB/c小鼠随机分为正常组(A组)、模型组(B组)、3个不同剂量给药组(C、D、E组)。除A组外其他组小鼠经2.0Gy60Coγ照射后的第3天进行腹腔注射环磷酰胺(CTX,50mg/kg),连续给药3d后完成骨髓抑制模型制备。小鼠造模后,第2天开始给药,每日每只小鼠灌胃量20ml/kg,A、B组灌入质量分数为0.9%生理氯化钠溶液,C、D、E组分别按四物汤颗粒剂的2.5、5.0、10.0g/kg溶于蒸馏水灌胃,共给药7d。应用IMDM培养基培养骨髓基质细胞,免疫印迹法观察各组对骨髓基质细胞TGF-β1和Smads信号通路表达的影响。结果①TGF-β1的表达:与A组(0.463±0.075)比较,B组骨髓基质细胞表达显著升高(1.284±0.043,P<0.05);与B组比较,C、D、E组可使TGF-β1表达降低;②Smads的表达:与A组(0.973±0.032)比较,B组骨髓基质细胞表达显著升高(2.036±0.044,P<0.05);与B组比较,C、D、E组可使Smads表达降低。结论四物汤配方颗粒可调节TGF-β1和Smads表达,对骨髓抑制小鼠造血微环境有一定改善。
Objective To observe the effect of Siwu Tang formula granule on hematogenesis function in bone marrow depressed mice and to explore its mechanism of hematopoietic regulation.Methods 50 mice were randomly divided into 5 groups,a normal group (group A),a model group (group B) and 3 treatment groups (group C,D,E).Siwu Tang was given by gavage to the mice in group C,D and E at the doses of 2.5,5.0 and 10.0 mg/kg (0.2 ml·10 g-1·d-1).The effects of the formula granule on TGF-β1/Smads signal transduction pathway of the bone marrow stromal cells in bone marrow depressed mice were tested by western blotting (WB).Results Compared with group A (0.463±0.075),the expression of TGF-β1 in group B of bone marrow stromal cells significantly increased (1.284±0.043,P0.05).Compared with group B,the expressions of TGF-β1 in group C,D,E significantly decreased.The expression of Smads of bone marrow stromal cells in group B (2.036±0.044) was higher than that of group A (0.973±0.032,P0.05).Compared with group B,the expressions of Smads in group C,D,E statistically decreased.Conclusion The results suggest that Siwu Tang formula granule may adjust TGF-β1/Smads to improve hematopoietic microenvironment.
