硝酸酯类一氧化氮供体药物研究进展

Development of nitrate NO-releasing agents

一氧化氮(nitric oxide,NO)具有多种生理活性,为了使经典药物发挥更广泛或者更好的治疗作用,研究人员将能释放NO的硝酸酯类基团与这些药物骈合,得到了一系新型硝酸酯类供体药物。这些药物涉及解热镇痛药、非甾体抗炎药、循环系统药和化学治疗药等。这类药物在体内被分解为母体药物和NO,二者分别发挥生理效应,达到协同增效作用。过去几年里,新型硝酸酯类NO供体药物受到了广泛研究,并取得了一些成果。文中综述了这类药物近年来的研究进展。

The physiological activity of nitric oxide （NO） is very extensive. For the purpose of developing more comprehensive or satisfactory therapy, these drugs are conjugated with the groups releasing NO, and a series of novel nitrate NO-releasing drugs have been obtained. Related classic drugs include antipyretic analgesics, non- steroidal anti-inflammatory drugs, circulator）, system drugs, and chemotherapeutic drugs. The new conjugated NO- releasing drugs are degraded into parent drug and NO in vivo. Both of them play their pharmacological effects to achieve synergy. During the past several years, this kind of new agents has been researched broadly, and some of them have succeeded. In this article, we reviewed the development of novel nitrate NO-releasing agents in recent years.
