血管内皮生长因子及中期因子在子宫内膜癌中的表达及意义 被引量:1

EXPRESSION AND SIGNIFICANCE OF VASCULAR ENDOTHELIAL GROWTH FACTOR AND MIDKINE IN ENDOMETRIAL CARCINOMA

摘要 目的探讨子宫内膜癌组织血管内皮生长因子(vascular endothelial growth factor,VEGF)和中期因子(midkine,MK)mRNA的表达水平及其相关性,分析二者与疾病进展的关系。方法应用实时荧光定量PCR,检测50例子宫内膜癌和30例正常子宫内膜组织中VEGF和MK mRNA的表达水平。结果子宫内膜癌组织中VEGF和MK mRNA表达量分别为28.61±10.08、10.16±2.19,明显高于正常对照组(P<0.05),且VEGF和MK的表达之间呈显著正相关(r=0.815,P<0.01)。VEGF和MK的表达在Ⅰ a和Ⅰ b期患者之间差异无统计学意义(P>0.05),在Ⅰ c和Ⅱ期患者的表达明显高于Ⅰa和Ⅰb期(P<0.05,P<0.01)。结论 VEGF、MK共同作用于子宫内膜癌的血管形成,且随恶性程度的进展表达逐渐增强,可作为评估子宫内膜癌恶性程度和不良预后的重要指标。 Objective To explore the expressions and correlation of vascular endothelial growth factor(VEGF) and midkine(MK) in endometrial carcinoma , and to analysis the relation with disease progression. Methods Real- time fluorescence quantitative PCR (FQ -PCR) was used to detect the expression of VEGF and MK mRNA in 5 0 endometrial carcinoma samples and 3 0 controls . Results VEGF and MK expression level in endometrial carcinoma samples were respectively 28.61 ± 10.08 and 10.16± 2.19, which significantly increased compared with control group ( P 〈 O. 05 ) . There was a negative linear correlation between VEGF and MK ( r = 0. 815, P 〈 0.01 ). The expression levels of VEGF and MK in stage I c and stage Ⅱ were significantly higher than those in stage I a and stage I b ( P 〈 0. 0 5 , P 〈 0. 0 1 ) , but there was no difference between stage I a and stage I b. Conclusion VEGF and MK have synergistic effect on angiogenesis, and the expression level gradually increased with tumor grade development. Over expressed VEGF and MK could be regarded as important parameters for estimating malignant degree and poor prognosis of endometrial carcinoma.
