摘要
目的探讨黄芪总黄酮对高糖诱导的牛视网膜血管周细胞氧化应激与DNA损伤的影响。方法体外培养3代融合的牛视网膜血管周细胞,分别在对照组、高糖组、黄芪总黄酮组中孵育6d,透射电镜观察周细胞超微结构改变,硫代巴比妥酸法检测培养液丙二醛(MDA)水平;黄嘌呤氧化酶反应系统检测培养液超氧化物歧化酶(SOD)水平;单细胞凝胶电泳检测DNA损伤情况。结果与高糖组相比,黄芪总黄酮组MDA/SOD比值降低;黄芪总黄酮组周细胞彗尾长度及拖尾率均减低。结论黄芪总黄酮对高糖培养下的牛视网膜血管周细胞氧化应激与DNA损伤有明显的抑制作用。
Objective:To study the effect of total flavonoids of astragalus on oxidative stress and DNA damage of bovine retinal perivascular cell under high glucose.Methods:3 passage cells were cultured for 6 days in the control group,the high glucose groups and the total flavonoids of astragalus group.The cell structure was observed under the transmission electron microscope.The contents of MDA were detected by the TBA method.The SOD level was used the xanthine oxidase method.The DNA damages were detected with the single cell gel electrophoresis.Results:Compared with the high glucose group,the ratio of MDA /SOD was decreased.The tail length and the comet rate were reduced in the total flavonoids of astragalus group.Conclusion:The oxidative stress and the DNA damage of bovine retinal perivascular cell under high glucose could be inhibited obviously by the total flavonoids of astragalus.
