摘要
纸层析、紫外-可见光谱和红外光谱法研究了紫荆花红色素的化学结构。结果表明:用展开剂BHW分离色素出现4个色斑,分别命名为组分1、2、3、4,其中组分1为红色,254nm紫外照射有黄色荧光;紫外可见光谱扫描,λmax-vis为531nm,加入AlCl3后Δλmax-vis为0,A440nm/Amax-vis为49%,在300~335nm存在吸收峰,440nm处未见肩峰;KBr压片,红外扫描观察谱图,主要波数为3439、2926、1632、1437、1385、1011、800、565cm-1。综上可推断紫荆花红色素为芍药色素,可能为芍药色素-3,5双葡萄糖,且分子内带有肉桂酸式酰基结构。
Paper chromatography,ultraviolet-visible spectrum and infrared spectrum methods were used to study the chemical structure of anthocyanin of red pigment from flowers of Cercis chinensis bge.The paper chromatography showed that BHW can separate the pigment well into four circular spots named 1、2、3、4 respectively.Component 1 is red and yellow fluorescence through UV irradiation is 254nm.The UV spectrum analysis showed that:In 0.1%HCl-MeOH solution,the wavelength of λmax-vis is 531nm and λmax-vis is 0 nm after adding 5% AlCl3;the absorbance ratio of A440nm/Amax-vis is 49%.An absorption peak(318nm)in the wavelength of 300~330nm was found,and no acromion near the wavelength of 440nm.IR spectrum revealed that the main absorption wave numbers are 3439、2926、1632、1437、1385、1011、800 and 565cm-1.The conclusion is that this red pigment is paeonidin,the structure may be paeonidin-3,5-glucose(Pn-3,5-glu)with cinnamic acid acyl structure.The sugar ligand types,connections of glycosidic bond and acylation structure are required for further studies.
