齿瓣石斛AP3基因的克隆及序列分析

摘要 采用RACE技术从石斛花序中克隆了MADS-box家族AP3基因,该基因全长947 bp,包括完整的编码区、5′-UTR和3′-UTR,编码227个氨基酸.生物信息学分析表明,该基因编码相对分子量为26.5 KD的碱性疏水蛋白,具有保守MADS区和半保守K区结构域,与蝴蝶兰和拟南芥AP3同源蛋白相似性高达92%和90%. The paper has cloned the AP3 gene of MADS box family from inflorescence of Dendrobium Devonianum by RACE.The gene is 947 bp in length,including the whole coding region,5-UTR and 3-UTR,and encodes a polypeptide consisting of 227 amino acid residues.The result of bioinformatics analysis shows that DdAP3 protein is a 26.5 KD hydrophobic alkality protein,and contains a conserved MADS and a conserved-half K-box domain.Moreover,its amino acids sequence shows 92% and 90% identity to the AP3 homology proteins of Phalaenopsis aphrodite and Arabidopsis thaliana respectively.
