摘要 目的探讨不同抗凝剂、不同保存温度等标本相关因素与全血葡萄糖-6-磷酸脱氢酶(G6PD)活性检测的相关性,确定G6PD活性检测的标本最适条件。方法采集血液标本20例、分别用枸橼酸葡萄糖(ACD)、枸橼酸钠和乙二胺四乙酸二钾(EDTA-K_2)3种抗凝剂抗凝,测定其G6PD活性及G6PD活性浓度;同一例血标本用ACD、枸橼酸钠和EDTA-K_2抗凝剂分别抗凝,分成2~8℃保存组及常温保存组,分别在第1~34天测定其G6PD活性浓度。结果 ACD、枸橼酸钠和EDTA-K_2 3种不同抗凝剂抗凝全血标本测得的G6PD活性差异有统计学意义(P<0.05),但G6PD酶活性浓度的检测结果差异无统计学意义(P>0.05);3种抗凝剂的标本在2~8℃保存条件下34 d内测得的G6PD活性浓度差异无统计学意义(P>0.05),标本在常温保存条件下,34 d内测得的G6PD活性浓度检测结果差异有统计学意义(P<0.05)。结论 ACD、枸橼酸钠和EDTA-K_2 3种抗凝剂抗凝全血标本用于G6PD活性浓度检测时,其检验结果具有良好的一致性;在2~8℃保存条件下,3种抗凝剂抗凝全血标本用于G6PD活性浓度检测可保存34 d。 Objective To analyze the factors which effect the activity of G6PD. Methods The G6PD activity and the G6PD concentration of 20 samples was detected. The blood samples were anticoagulated by citrate dextrose(ACD),natrium citricum and EDTA-K2 ; One sample with different anticoagulant of citrate dextrose(ACD), natrium citricum and EDTA K2 were stored at 2 -8 ℃ and room temperature for 34 days, The G6PD concentration of enzymatic was detected each day from 1-34 days. Results There were significant difference of the G6PD activity in different groups anticoagulated by ACD,natrium citrieum and EDTA-K2 (P〈0.05). There was no significant difference of the G6PD concentration of enzymatic activity between different groups anticoagulated by ACf),natrium citricum and EDTA-K2 (P〉0.05). No significant difference of the G6PD concentration of enzymatic activity between different groups anticoagulated by ACD,natrium citricum and EDTA-Ke(P〈0. 05)stored at 2-8 ℃ for 34 days(P〉 0.05). Significant difference of the G6PD concentration of enzymatic activity between different groups antieoagulated by ACD,natrium eitricum and EDTA-K2 stored at room temperature for 34 days(P〈0.05). Conclusion The blood sample for G6PD concentration of enzymatic activity can be anticoagulated by ACD,natrium citricum and EDTA K2 and stored at 2-8 ℃ for 34 days.
