摘要
目的:建立以高效液相色谱法测定奥沙利铂脂质体中奥沙利铂含量的方法。方法:色谱柱为Hypersil C18(250 mm×4.6mm,5μm),流动相为甲醇:水=5∶95(V/V),流速为1.0 mL·min-1,检测波长为250 nm,柱温为30℃。结果:奥沙利铂的检测浓度在12.2~122.0μg·mL-1范围内线性关系良好;以峰面积(Y)为对浓度(C)进行回归分析,得回归方程为Y=78 575 C-4 811.8(n=5,r=0.999 9),奥沙利铂的检测浓度在12.2~122.0μg·mL-1范围内线性关系良好;检测限为97.6 ng·mL-1;平均回收率为98.58%,RSD=1.83%。结论:该方法操作简便、灵敏、准确,可用于奥沙利铂脂质体中奥沙利铂的含量测定。
OBJECTIVE:To establish an HPLC method for the content determination of oxaliplatin in Oxaliplatin liposome. METHODS:The separation was performed on Hypersil C18(250 mm×4.6 mm,5 μm) column with methanol-water(5 ∶ 95,V/V) as mobile phase at flow rate of 1.0 mL·min^-1. The detection wavelength was set at 250 nm and column temperature was 30 ℃. RESULTS:The linear range of oxaliplatin was 12.2~122.0 μg.mL^-1(n=5,r=0.999 9) with an average recovery of 98.58%(RSD=1.83%). The limit of detection(LOD) was 97.6 ng·mL^-1. CONCLUSION:The method is simple,sensitive and accurate for the content determination of oxaliplatin in Oxaliplatin liposome.
