摘要
对大鼠肾小球系膜细胞(GMC)进行体外培养,利用单克隆抗体进行间接免疫荧光染色,结果抗Vi-mentin和抗Desmin抗体均为阳性,而抗Keratin抗体为阴性,证实培养的细胞为纯的GMc;并探索了GMC培养的最佳条件,且对肾小球的收集方法作了改进,结果显示,该方法收集的肾小球数多,肾小土脱落彻底,且肾小球贴壁率高,使得GMC生长既多又好。
This article concentrated on the method for culturing rat glomerular mesangial cells (GMCs) in vitro.GMCs were identified by indirect immunofluorescent staining with monoclonal antibody. The results showed that the staining using anti-vimentin and anti -desmin antibody were positive reaction, anti-keratin were negative,which indicated that the cultured cells were quite pure GMCs. The best condition in culturing GMCs and improvement of the method for collecting glomeruli were also investigated. The study has indicated that the approach can provide a sufficient numbers of glomeruli Bowman's Capsules come off thoroughly and the rate of attaching wall of GMCs is significantly increased. These in vitro observations may demonstrate that GMCs grow better and more rapidly.
