摘要 目的:建立一种简便、快捷、准确的血清α-L-岩藻糖苷酶检测方法。方法;报道了血清α-L-岩藻糖苷酶动力学连续监测法。结果:α-L-岩藻糖苷酶对硝基苯α-L-岩藻吡喃喝糖苷的米氏常数为0.133±0.052mmol/L,最适pH为5.0,延迟时间30s,线性反应时间从60~180s,批内CV5.2%,批间CV6.99%。该法基本解决了血清黄疸与微量溶血等因数对测定结果的影响。新疆地区的α-L-岩藻糖苷酶活性正常参考值范围为96.25±81.8nkat/L(2s),肝癌患者AFU阳性率为76.0%(19/25)。结论:研究结果表明,该法具有简单、快速、灵敏度高、准确等优点,适合于自动化检测和临床应用。 Objective To establish a simple , rapid and accurate serumα - L -Fucosidase (AFU)testing method.Methods A continuous monitoring method of the Dynamic Determination for Serum AFU. Result P-nitropheny α-L-Fuco-pyranoside the Km's is the 0. 133±0.052 mmol/L. The most appropriate PH is 5. 0 and the delay time was 30 seconds. The time of linear reaction lasts from 60 seconds to 180 seconds. The intra-assay CV is 5. 2% and inter-assay CV is 6. 99 %. The method basically solves the affection to the testing results from the factor such as serum jaundice and micro - haemolysis. The reference HFU value of normal person in Xinjiang area is 96. 25±81. 8 nkat/L (2s). The AFU positive rate of hepatocellular carcinoma is 76. 0% (19/25). Conclusion The result shows that the method is rapid, simple, accurate, and sensitive. It is suitable for the automatic assay and clinical application.
