活血益气方对心梗后大鼠缺血心肌血管新生及VEGF表达的影响 被引量:16

Effects of Qi supplement and Blood Activation prescription on angiogenesis and VEGF expression in ischemic myocardium in rats model after myocardial infarction

摘要 目的:观察活血益气方对心肌梗死后大鼠缺血心肌血管新生的作用及其相关生长因子VEGF表达的影响。方法:健康雄性SD大鼠行左冠状动脉前降支结扎术致急性心肌梗死模型,随机分为模型组、活血益气方组和倍他乐克组,并设立假手术组,假手术组只穿线不结扎。连续给予相应药物4周后,处死大鼠,迅速摘取心脏在乳头肌横切面切取心肌组织,4%的多聚甲醛固定,石蜡包埋,切片。以免疫组化法测定心梗边缘区微血管数和VEGF的表达;取梗塞边缘区心肌组织,进行总RNA抽提,RT-PCR法检测心梗边缘区VEGF mRNA的表达。结果:(1)假手术组、模型组、倍他乐克组和活血益气方组均可看到新生的微血管。假手术组可见微血管增生不明显,模型组可见少量微血管增生,倍他乐克组和活血益气方组可见较多增生的微血管。模型组与假手术组比较,有统计学差异;倍他乐克组和活血益气方组与模型组比较,有统计学差异;(2)各组大鼠梗塞边缘区均可见到VEGF表达,模型组VEGF表达高于假手术组(P<0.05),倍他乐克组和活血益气方组VEGF表达高于模型组(P<0.01);(3)模型组VEGF mRNA表达水平高于假手术组(P<0.05),活血益气方组VEGF mRNA表达高于模型组(P<0.05)。结论:活血益气方对心肌梗死后大鼠缺血心肌血管新生具有促进作用,其作用机制可能与上调心肌局部VEGF及其mRNA表达有关。 Objective:To observe the effects of Qi supplement and Blood Activation prescription on angiogenesis and one of its relative growth factor VEGF in ischemic myocardium in rats model after myocardial infarction.Methods:A model of acute myocardial infarction was constructed through ligating the ramus descendens anterior arteriae coronariae sinistrae in SD rats.All the model rats were divided into model group,Qi supplement and blood activation prescription group,and Metoprolol group randomly.The sham-operation group was established as controls.The rats were killed after treat with corresponding drugs for 4 weeks.The cardiac muscle were fixed by 4% Polyoxymethylene,and paraffin imbedding.The micro vessel count and expression of VEGF protein in marginal zone of infracted region were measured by immunohistochemical method.The total RNA was isolated from infracted region of left ventricular myocardium and the same regions of control group.The expression of VEGF mRNA was measured by Reverse Transcription-Polymerase Chain Reaction.Results:(1) There is a little capillary vessel proliferation in the sham operated group.The capillary vessel proliferation in the model group is more than in the sham operated group(P 0.01).The capillary vessel proliferation in the (P 0.01).(2)The expression of VEGF protein in the model group is higher than the sham operated group (P 0.05),the expression of VEGF protein in the Qi supplement and blood activation prescription group and Metoprolol group is higher than the model group(P 0.01).(3)The expression of VEGF mRNA in the model group is higher than the sham operated group (P 0.05),the expression of VEGF mRNA in the Qi supplement and blood activation prescription group is higher than the model group (P 0.05).Conclusion Qi supplement and blood activation prescription have a role in promoting angiogenesis in ischemia myocardial after myocardial infarction.The mechanism may be related to increase the expression of VEGF and its mRNA in regional myocardial.Conclusion:
