涤痰汤对小鼠学习记忆功能障碍的影响 被引量:3

摘要 目的探讨涤痰汤对小鼠记忆功能障碍的影响及其可能的机制。方法用东莨菪碱、亚硝酸钠和乙醇分别建立记忆获得障碍、记忆巩固障碍和记忆再现障碍模型,通过跳台法观察涤痰汤对记忆障碍小鼠学习记忆三个环节的影响。采用免疫组织化学法检测皮质区乙酰胆碱酯酶(AchE)、乙酰胆碱转移酶(ChAT)的表达;使用高效液相色谱法测定脑组织内多巴胺(DA)的含量。结果跳台实验:三种模型组在测试中,错误次数都较正常组明显增加,而潜伏期较正常组明显缩短(P<0.01);涤痰汤可不同程度的改善模型小鼠的学习记忆获得、巩固、再现障碍,与模型组比较其差异有统计学意义。指标检测:涤痰汤组AchE的表达明显降低,ChAT的表达增加,与模型组比较其差异均有统计学意义(P<0.05);涤痰汤可以明显抑制DA含量的下降,与模型组比较其差异有统计学意义(P<0.01)。结论涤痰汤对学习记忆有改善作用,其机制可能与降低脑内AchE的表达、提高ChAT的表达、抑制DA含量的下降相关。 Objective To study the effects of Ditan Decoction on abilities of learning and memory and its preliminary mechanisms.Methods The methods of platform were used to investigate the effects of Ditan Decoction on the dementia models challenged by scopolamine hydrobromide,alcohol,NaNO2 in Kunming mice.The expression of acetylcholine esterase(AchE) and acetylcholine transferase(ChAT) in hippocampus were measured by immunohistochemical method.Further we measured the contents of dopamine(DA) in brain tissue by High Performance Liquid Chromatograph.Results Compared with normal group in the test of platform,error times increased significantly in three model groups,in which the incubation period was significantly shorter(P 0.01).Ditan Decoction can improve memory impairment induced by scopolamine,alcohol and NaNO2 to some extent.Compared with the model group,Ditan Decoction could significantly decrease the expression of AchE and increase the expression of ChAT(P 0.05)and could significantly inhibit the decline of DA content in brain tissue(P 0.01).Conclusion Ditan Decoction could improve the abilities of learning and memory in mice models through multiple ways.The preliminary mechanisms may be associated with its effects on decrasing the expression of AchE,increasing the expression of ChAT and inhibit the decline of DA content in brain tissue.
