The expression and the clinical implication of CD87 on acute myeloid leukemia cells in children

摘要 目的探讨小儿急性髓性白血病细胞尿激酶受体(uPAR,CD87)表达及其临床意义。方法应用流式细胞术(FCM)检测了54例急性髓性白血病(AML)及20例急性淋巴细胞白血病(ALL)患儿CD87的表达,还用FCM及逆转录聚合酶链反应(RTPCR)法检测了U937、HL60、K562细胞在氟波酯(PMA)作用下诱导分化过程中CD87表达的改变。结果54例AML中33例(61%)CD87为阳性表达,20例ALL均为阴性表达。CD87阳性组与阴性组AML发生髓外浸润的比例分别为88%和19%(P<0001);外周血幼稚细胞百分率分别为(610±215)%和(305±152)%(P<0001);完全缓解(CR)率分别为52%和86%(P<005);发生出血的比例分别为73%和62%(P>005)。U937、HL60、K562经PMA诱导分化后,CD87表达增高。结论CD87可作为白血病诊断、分型、判断预后的指标之一,也可作为诱导分化的指标之一。 Objective To explore the expression of receptor for urokinase (CD87) on acute myeloid leukemia (AML) cells in children and its clinical implication Methods The expression of CD87 was detected with flow cytometry (FCM) in 54 AML patients and 20 acute lymphoblastic leukemia (ALL) patients The expressionsa of CD87 on U 937, HL60 and K562 cells were also studied with FCM and reverse transcriptionpolymerase chain reaction (RTPCR) during differentiation induced by phorbol myristate acetate (PMA) in vitro Results The staining for CD87 was positive in 33 of 54 patients with AML (61%) and negative in 20 patients with ALL In CD87 positive and negative groups of AML, the percentages of infiltration of extramedullary tissues were 88% and 19% (P<0001), respectively The percentages of blast cells in peripheral white blood cells were(610215)% and(305152)% (P<0001), respectively The complete remission rates were 52% and 86% (P<005), respectively The percentages of bleeding tendency were 73% and 62% (P>005), respectively The CD87 expressions on U937, HL60 and K562 cells were increasing during differentiation induced by PMA Conclusion CD87 might be used as one of the markers for leukemia diagnosis, typing and prognosis, and also for myeloid leukemia cells differentiation
