摘要
针对高校生物科学实验教学中存在的问题,着重探究了基因重组试验中的α-互补与蓝白斑筛选机制,精心设计了5组试验,结果指出,经双酶切的质粒载体自身不能环化,也不能成功转化;蓝白斑筛选中的蓝斑通常来源于商品载体中的环状质粒或者线性载体自连后的转化产物;重组子一般为白斑,但在一定条件下,白斑也会变成蓝斑。在此基础上作了进一步的讨论,为今后的教学实践和科学研究提供借鉴和指导。
Based on the questions of biological experiments in college teaching,the research explored the mechanism of α-complementation and blue-white screening and precisely designed 5 experiments.The results showed that plasmid vector could not form loop itself or transform successfully.The blue patch can be commonly from circular plasmid or transformation products of self-ligation.The recombinants usually display white patches,but the white patches can become blue under specific conditions.The results provided directions for future research and teaching practice.
