用蚀斑形成试验分离虫媒病毒

摘要 本文报告用蚀斑形成试验分离虫媒病毒。从643个蚊虫混合标本中分离到乙脑病毒104株,甲病毒1株。经过C6/36白纹伊蚊细胞增殖后,标本阳性率为14.2%,直接从蚊虫混合标本分离的阳性率为13.2%,前者的蚀斑数显著高于后者(P<0.001)。乙脑病毒蚀斑于接种后第4~9天出现,直径约6mm,边缘较模糊。甲病毒蚀斑于接种后第2~3天出现,直径1~2mm,较乙脑病毒小。用蚀斑形成试验分离虫媒病毒的优点是可以根据蚀斑的形态和出现时间,初步区别分离物是甲病毒属或黄病毒属。 A system for isolation of arboviruses is reported. 104 strains of JEV and 1 alphavirus strain wereisolated from 643 mosquitoe pools by plaque assay.Plaque assays after multiplication in 06/36 cell line, produced 14.2% positive results, as compared to 13.2% of the original inoculum. Besides, the number of plaque formation in the fomer was significantly greater(P<0.001)than in the later.JEV plaques appeared in 4-9 days postinooulation, were hazy-tooking and about 6 mm in diameter, alpha virus plaques appeared 2-3days postinooulation and were 2mm in diameter.the plaque assay is advantageous in that, it provide preliminary differentiation of alphaviruses from flavaviruses.
