摘要
目的观察二苯乙烯苷(TSG)对SAMP8鼠脑组织胆碱能神经递质及单胺类神经递质含量的影响。方法选用6月龄的SAMP8鼠50只,随机分为5组,SAMP8对照组、阳性药石杉碱甲对照组、TSG低、中、高剂量组,每组10只。另选用6月龄SAMR1鼠10只作正常对照组。各组分别灌胃相应药物50天后,采用分光光度法检测各组小鼠脑组织胆碱乙酰转移酶(ChAT)和乙酰胆碱酯酶(AchE)活性,高效液相色谱法检测各组小鼠脑组织去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)含量。结果 SAMP8对照组脑组织ChAT活性明显降低,AchE活性明显升高,NE、DA、5-HT的含量明显降低,与正常对照组比较有显著性差异(P<0.05或0.01);TSG中、高剂量组ChAT活性显著升高,TSG各剂量组AchE活性显著降低(P<0.05或0.01),NE、DA、5-HT含量均得到提高,与SAMP8对照组比较,差异具有显著性(P<0.05或0.01)。结论二苯乙烯苷可有效促进乙酰胆碱(Ach)的合成及减少乙酰胆碱的分解,促进单胺类神经递质的合成,具有良好的抗痴呆的作用。
Objective To observe the effect of TSG on cholinergic neurotransmitter and monoamine neurotransmitter contents in SAMP8 mice's brain.Methods 50 mice of 6 month's old with SAMP8 were randomized divided to 5 groups(10 in each group):SAMP8 group,huperzine A control group,low-,mid-and high-dose groups of TSG.Another 10 mice were assigned to normal control group.After medication for 50 days,the activity of ChAT,AchE in mice's brain in each group were detected by spectrophotometry.The NE,DA and 5-HT contents in brain were detected by HPLC.Results The activity of ChAT decreased and the activity of AchE enhanced significantly,and the NE,DA and 5-HT contents decreased significantly in SAMP8 mice's brain comparing with normal group(P〈0.05 or 0.01).In TSG-treatment group,the activity of ChAT enhanced and the activity of AchE decreased significantly,and the NE,DA and 5-HT contents increased significantly in mice's brain comparing with SAMP8 group(P〈0.05 or 0.01).Conclusion TSG can effectively promote the synthesis of acetylcholine and reduce the decomposition of acetylcholine,promote the synthesis of monoamine neurotransmitter,resulting in the satisfactory anti-dementia effects.
