草鱼胆中毒大鼠致急性肾损伤TNF-α动态水平测定

摘要 目的:研究血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)在不同时间段鱼胆中毒大鼠体内的变化及其与鱼胆中毒后急性肾损伤(acute kidney injury,AKI)发生和发展的关系。方法:健康SD大鼠30只(雌雄各半),采用ELISA测定20只不同时间段鱼胆中毒大鼠与10只正常对照大鼠的血清TNF-α水平,比较鱼胆中毒大鼠血清TNF-α与正常对照大鼠血清TNF-α的水平。结果:①草鱼胆中毒大鼠肾组织病理改变主要在肾小管,表现为变性、坏死并且随时间延长病变越严重;②鱼胆中毒大鼠血TNF-α较正常对照组明显升高,其升高程度与时间成正比。结论:鱼胆中毒的发病机制与血清TNF-α密切相关,血清TNF-α参与了鱼胆中毒后AKI的发生与进展。鱼胆中毒时间越长引起中毒的程度越重,血清TNF-α升高就越明显。 Objective:To Investigate the relation of serum tumor necrosis factor-α(TNF-α)change in rats with different periods of fish bile poisoning and the occurrence,progress of acute kidney injury(AKI)after fish bile poisoning.Methods:Taking 30 SD rats as the subjects,using ELISA to detect the dynamic level of serum TNF-α in 20 rats with different periods of fish bile poisoning and 10 normal control rats.Then the serum TNF-α level of two groups were compared Results:(1) Renal pathological changes mainly occurred in the tubules caused by fish bile poisoning,and showed degeneration and necrosis,as well as more severe lesions with time(.2)Fish bile poisoning in rat serum TNF-α was significantly higher than the normal control group,the increase is proportional to the degree and time.Conclusion:The pathogenesis of fish bile poisoning has close relation with the serum TNF-α.The serum TNF-α has participated in the morbidity of fish bile poisoning,the occurrence and advancement of AKI after fish bile poisoning.The longer of fish bile poisoning,the more serious of poisoning degree,and the serum TNF-α level increase more obviously.
