摘要
利用本实验室筛选得到的苯乳酸生产菌Lactobacillus plantarum P421进行高产菌株的诱变选育。通过紫外线诱变进行诱变育种,以产物耐受和抑菌圈相结合的筛选方法,获得了一株高产苯乳酸的优良突变菌株,其产量达到528mg/L,比出发菌株81mg/L提高了6.5倍,且遗传性质稳定。该诱变方法和筛选方法目的明确,易操作,对类似的诱变育种具有一定的借鉴意义。
Phenyllactic acid-producing strain Lactobacillus plantarum P421 which screened by our lab was used as starting strain. The mutation of ultraviolet on L.plantarum P421 was conducted,and the fast screening method based on phenyllactic acid tolerance and inhibition zone were established. After mutation,the phenyllactic acid production ability was increased to 528mg /L,which was 6.5 times than that of the original strain of 81mg /L,and the heritability was stable. The purpose of mutation and screening methods are clear and easy to operate which provide reference to similar mutation breeding.
