摘要
目的应用转染了Bcl-2基因的鼠T-细胞淋巴瘤WHb12细胞系进一步研究Bcl-2基因的抗凋亡作用及意义。方法分别采用紫外线照射20s、10μm地塞米松作用24h和200ng/ml鬼臼乙叉甙作用24h等方法,诱导WHb12细胞和其亲本细胞W72USA细胞凋亡,与亲本细胞相比较,观察转染了Bcl-2基因的WHb12细胞是否具有抗凋亡作用。结果未转染Bcl-2基因的亲本细胞W72USA细胞发生了凋亡,而转染了Bcl-2基因的WHb12细胞存活完好,无明显凋亡发生。结论进一步证实了Bcl-2基因具有显著的抗凋亡作用,其抗凋亡作用谱广,具有重要意义。
Objective The effect and significance of Bcl-2 gene on apoptosis were investigated using WHb 12 cell line with transferred Bcl-2 gene from murine T-cell lymphoma.Methods The apoptosis was induced by UV radiation for 20 seconds,10 μm dexamethasone and 200 ng/ml etopside for 24 hours separately,in the WHb 12 cells and their parental W 7.2 USA cells for comparison to observe whether the WHb 12 cells with transferred Bcl-2 gene are the apoptotic blocker.Results It is showed that the apoptosis occurr in the parental W 7.2 USA cells without transfererd Bcl-2 gene,whereas it did not occurred in the WHb 12 cells with transferred Bcl-2 gene,which survived very well.Conclusion This result comfirmed that Bcl-2 gene is a powerful and broad spectrum blocker of apoptosis.
