摘要
以魁蚶血浆为原料,经磷酸缓冲液抽提、75%饱和度硫酸铵沉淀、DE52纤维素离子交换层析和Sephadex G-100分子筛层析等步骤,获得了魁蚶防卫素。经SDS-PAGE检测,魁蚶防卫素分子量为76·2kDa。对魁蚶防卫素进行抗菌活性检测发现,魁蚶防卫素对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、变形杆菌和产气杆菌均有较强的抑制作用。
A defensin(SBD)was isolated from Scapharca broughtonii by precipitation with 75% saturation of(NH4)2SO4,ion-exchange on DE52 cellulose and gel filtration on Sephadex G-100.SDS-PAGE showed that SBD had a molecular mass of 76.2kDa.The antibacterial activity test showed that SBD had strong inhibiting effect to Staphylococcus aureus,Bacillus subtilis,Escherichia coli,Protrus vularis and Enterobacter aerogenes.
