摘要
目的:研究粉防己碱(tetrandrine,Tet)对脂多糖(lipopolysacchride,LPS)诱导的雄性Sprague-Dawley(SD)大鼠发热模型的解热作用及其机制。方法:48只雄性SD大鼠随机分为空白对照组、LPS模型组、对乙酰氨基酚对照组及粉防己碱5,15,45mg/kg组。用LPS100μg/kg腹腔注射建立大鼠发热模型,药物处理后每30min测体温一次,连续监测6h,观察大鼠的体温变化;运用Western-blot检测脑组织中环加氧酶2(Cyclooxygenase-2,COX-2)的表达,并用EIA法检测其前列腺素E2(Prostaglandin E2,PGE2)的含量;髓过氧化物酶(Myeloperoxidase,MPO)试剂盒检测肝组织MPO水平。结果:粉防己碱能有效抑制LPS诱导的大鼠发热反应,显著降低脑组织中COX-2和PGE2的表达,抑制肝脏MPO水平。结论:粉防己碱具有显著的解热作用,其作用机制可能与抑制脑组织中COX-2和PGE2的表达,降低肝组织中MPO含量有关。
AIM: To observe the effect of tetrandrine on LPS-induced febrile response in sprague-dawley(SD) rats and elucidate the antipyretic mechanism of tetrandrine. METHODS: Forty eight male SD rats were randomly assigned to 6 groups: tetrandrine 5,15,45 mg/kg group,model group,normal group,acetaminophen 50 mg/kg positive control group. Fever was induced by intraperitoneally injecting LPS (100 μg/kg),and the temperature was observed every 30 minutes,the observation lasted for 6 h,we observed the changes of temperature. Western blotting technologies and euzyme-linked immunosorbent assays were applied to the expression of cyclooxygenase-2 (COX-2) and Prostaglandin E2(PGE2) in rat brain tissues,and myeloperoxidase(MPO) in liver was tested by special kits. RESULTS: Tetrandrine could effectively decrease febrile response,inhibit the expression of COX-2 and PGE2 in brain,lower liver MPO level in LPS-induced fever rats. CONCLUSION: Tetrandrine has the action of antipyretic effect. Its mechanism may be related to inhibition of the expression of COX-2 and PGE2 in brain tissues,and level of MPO in liver.
