涤痰汤对阿尔茨海默病模型大鼠行为学影响的实验研究

Experimental study on effect of Ditan decoction on ethology in model rats with Alzheimer's disease

目的观察涤痰汤对阿尔茨海默病(AD)模型大鼠行为学的影响。方法将大鼠随机分为正常组(A组)、假手术组(B组)、模型组(C组)、哈伯因组(D组)和涤痰汤组(E组),采用大鼠前脑基底核注射Aβ25-35和鹅膏覃氨酸混合液诱导复合AD动物模型,E组和D组灌胃给药28 d。用Morris水迷宫系统测定各组的逃避潜伏期及跨台次数。结果 C组大鼠逃避潜伏期、平均游泳距离、跨台次数与A组、D组、E组比较均有显著性差异。在定位航行试验中,E组前2 d逃避潜伏期、40%区域较D组明显减少;在空间搜索试验中,E组20%区域少于D组。其余指标2组间均无明显差异。结论涤痰汤对于AD模型大鼠的空间学习记忆功能受损具有明显保护作用,且涤痰汤的保护作用稍优于哈伯因。

ObjectiveIt is to observe the effect of Ditan decoction on ethology in model rats with Alzheimer＇s disease（AD）.MethodsThe Rats were randomly divided into normal group（group A）, sham operation group（group B）,model group（group C）,Huperzine A-Zhulin Antun group（group D）,Ditan decoction group（group E）.AD animal models of rats were made by injecting mixed liquor of Aβ25-35 and IBO in their forebrain basal nucleus.Latent period of escape and times of bestriding berm in every group were detected by Morris water maze system after 28 days＇ treatment by intragastric administration in group E and group D.ResultsThe difference in the rats＇ ability of escape latent period,average swim length,times of bestriding berm in group C was significant compared with group A,group D and group E.In the position moving test,escape latent period and 40% area in 2 days in group E was significantly less than that in group D.In space searching test,20% area in group E was less than that in group D.There was no significant difference in other indexes between the two groups.Conclusion Ditan decoction has a obvious protecting function on the space learning and memory ability of rat models with AD,and its protecting effect is a little better than Huperzine A-Zhulin Antun.
