摘要
目的观察姜黄素对佐剂性关节炎大鼠成纤维样滑膜细胞(FLS)释放血管内皮生长因子(VEGF)、白细胞介素-6(IL-6)的影响。方法体外原代培养佐剂性关节炎大鼠FLS。取第3代FLS分别加入浓度为0,10,20,40,80μmol/L的姜黄素溶液,共同培养24h,48 h,72 h。ELISA法检测共培养上清液中VEGF、IL-6含量。结果共培养24 h后40,80μmol/L浓度组和48 h、72 h后所有浓度组的VEGF值与相应空白对照组比较均有显著性差异。共培养24 h后40,80μmol/L浓度组,48 h后20,40,80μmol/L浓度组和72 h后所有浓度组IL-6值相对对照组均明显降低。结论姜黄素能抑制佐剂性关节炎大鼠成纤维样滑膜细胞释放VEGF、IL-6,这可能是姜黄素治疗类风湿关节炎的作用机制之一。
Objective It is to observe the effect of curcumin on the secretion of vascular endothelial growth factor(VEGF) and interleukin-6(IL-6) by the fibroblast-like synoviocyte(FLS) in adjuvant arthritis rats.Methods FLS was primary cultured synoviocytes from adjuvant arthritis rats in vitro,and the third generation was added with 0,10,20,40 and 80 μmol/L curcumin to co-culture for 24 h,48 h and 72 h.ELISA method was used to determine the concentration of VEGF and IL-6 in clear supernatant liquid after co-culture.Results After co-cultured for 24 h,VEGF in 40,80 μmol/L curcumin groups and all different levels of curcumin groups after 48 h,72 h was significant different compared with correlated control group.The value of IL-6 of 40 and 80 μmol/L curcumin groups after co-cultured for 24 h,20,40 and 80 μmol/L curcumin groups after 48 h,and all groups after 72 h all obviously decreased,compared to the blank group.ConclusionCurcumin can restraint the secretion of VEGF and IL-6 by FLS in adjuvant arthritis rats,which may be the mechanism of the treatment for rheumatoid arthritis with Curcumin.
