摘要
以红富士（ＶｉｔｉｓｖｉｎｉｆｅｒａＬ．×Ｖ．ｌａｂｒｕｓｃａＬ．）葡萄果实为试材，以水为脱落酸（ＡＢＡ）的浸提剂，利用ＡＢＡ单克隆抗体ＭＡＣ６２，建立了一套适合葡萄果实微量样品，且样品浸提液不需进一步纯化就可直接用于ＡＢＡ测定的放射免疫测定技术（ＲＩＡ）。把ＲＩＡ法与高效液相色谱法（ＨＰＬＣ）进行了比较，ＲＩＡ以其简便快捷，灵敏度高，专一性强，测得结果重复性好为显著特点，显示出比ＨＰＬＣ更大的优越性。此方法也可在果树其它器官上应用。
This experiment was conducted with fruits of Hongfushi grapevine( Vitis vinifera L.× V.labrusca L.).Using the monoclonal antibody of abscisic acid(ABA)and double distilled water as the extracting solvent for the extraction of ABA from tissues of grape berry,a newly developed radioimmunoassay(RIA)for directly measuring ABA in crude extracts of grape berry was described.RIA and high performance liquid chromatography(HPLC)for measuring ABA in grape berry were compared.RIA was proved to be more sensitive,specific,precise and efficient.
