氚标记甲基-苯基-四氢吡啶在鼠脑内的结合部位——帕金森病发病机理的研究

摘要 本实验运用受体放射自显影术观察神经毒素甲基-苯基-四氢吡啶(MPTP)在大鼠脑内的分布,并运用亚细胞成份分离技术研究MPTP在小鼠脑亚细胞成分中的分布。结果表明,~3H—MPTP在大鼠脑内有其特异性的结合位点,这一位点能被非标记的MPTP和单胺氧化酶抑制剂优降宁竞争结合。~3H-MPTP亚细胞的分布显示,该物质与线粒体的结合量最大。以上结果提示,~3H—MPTP在脑内的受体可能为单胺氧化酶,该酶在帕金森病的发病机理中有可能起着某种重要的作用。 The distribution of methyl-phenyl-tetrahydropyridine (MPTP), a neurotoxin, in rat brain and organellae of mouse brain was studied by receptor autoradiography and subcellular fractionation respectively. The studies demonstrated that in rat brain there existed specific ~3H-MPTP binding sites, which could be bound competitively by large amount of unlabeled MPTP and pargyline. The study of subcellular distribution showed that the highest binding amount of the agent was in mitochondrial fraction. The results suggest that the receptor of ~3H-MPTP in brain may be the monoamine oxidase, which might play an important role in the pathogenesis of Parkinson's disease.
