趋化因子受体CCR7在胃癌组织和细胞株中过度表达

摘要 趋化因子受体CCR7高表达与肿瘤淋巴结转移等因素密切相关.目的:检测CCR7在胃癌组织和细胞株中的表达情况,探讨CCR7在胃癌中的作用.方法:采用实时PCR技术检测30例胃癌组织及其配对癌旁组织中的CCR7 mRNA表达,RT-PCR检测人胃癌细胞株SGC-7901和AGS中的CCR7 mRNA表达.分析胃癌组织中的CCR7mRNA表达与胃癌临床病理特征间的相关性.结果:CCR7 mRNA在胃癌组织中的表达水平显著高于其配对癌旁组织(P〈0.05).人胃癌细胞株SGC-7901和AGS中均存在明显CCR7 mRNA表达.CCR7 mRNA表达与胃癌浸润深度、淋巴结转移和TNM分期相关(P〈0.05).结论:CCR7在胃癌中存在过度表达,可能参与了胃癌的进展过程. Background: Overexpression of chemokine receptor CCR7 is correlated with lymph node metastasis of cancers. Aims: To investigate the expression of CCR7 in gastric cancer tissue and cell lines, and to study its role in gastric cancer. Methods: Real-time PCR was used to detect the expression of CCR7 mRNA in cancer tissue and matched paraneoplastic tissue of 30 gastric cancer patients. RT-PCR was used to determine the expression of CCR7 mRNA in human gastric cancer cell lines SGC-7901 and AGS. The correlation between expression of CCR7 mRNA and the clinicopathologic characteristics of gastric cancer was analyzed. Results: Expression of CCR7 mRNA was significantly higher in gastric cancer tissue than that in matched paraneoplastic tissue (P〈0.05). CCR7 mRNA was expressed obviously in human gastric cancer cell lines SGC-7901 and AGS. CCR7 mRNA expression was correlated significantly with the invasiveness, lymph node metastasis and TNM staging of gastric cancer (P〈0.05). Conclusions: CCR7 is overexpressed in gastric cancer and might be involved in its progression.
