摘要
为探讨Fas在内毒素(ET)致大鼠肝细胞凋亡中的作用,将48只SD大鼠随机分为2组,1组为对照组(静脉注射生理盐水),2组为内毒素组(静脉注射内毒素5mg/kg)。分别在注射后第3、4、8、12h每组各剖杀6只大鼠,采取肝,用流式细胞术检测肝细胞凋亡情况,用免疫组织化学技术观察内毒素对肝细胞Fas表达的影响。结果显示,2组的肝细胞凋亡率明显高于1组(P<0.01),且随着时间的延长呈上升趋势;2组的Fas积分光密度明显高于1组(P<0.01),且一直呈上升趋势。结果表明,ET通过上调肝细胞中Fas的表达,可显著介导肝细胞的凋亡。
Forty-eight SD rats were randomly divided into two groups of the normal control group ino-culated with normal saline by intravenous injection and the endotoxin(ET) treatment group inoculated with ET at 5 mg/kg by intravenous injection to explore the effect of Fas on ET-induced hepatocyte apoptosis in rats. The liver samples were collected from 6 rats in each group at hour 3,4,8 and 12 post-injection, respec-tively. The apoptotie hepatocytes from the liver samples were examined by flow cytometry,and the effect of ET on the expression of Fas in the liver samples was observed by immunohistochemical examination. Com-pared with that in the control group, the hepatocyte percentage of apoptosis and IOD of Fas in the ET treatment group increased significantly(P〈0.01) along with prolongation of time after the injection. The results indicated that ET could stimulate the hepatocyte apoptosis through up-regulating the expression of Fas gene in liver.
