摘要 观察高三尖杉酯碱(HH)对人皮肤瘢痕组织成纤维细胞增殖的影响。利用MTT比色分析法及流式细胞术检测不同浓度HH(μg/ml)作用不同时间对体外培养的人皮肤瘢痕组织成纤维细胞增殖的影响。当HH浓度为0.12μg/ml时即可抑制体外培养的人皮肤瘢痕组织成纤维细胞的生长,0.66μg/ml时即可产生50%的抑制效应,10μg/ml时可完全抑制成纤维细胞的生长,且抑制率随时间的延长而增强,而对正常皮肤组织的成纤维细胞无作用。HH可抑制体外瘢痕组织成纤维细胞的增殖且抑制作用呈剂量时间依赖性。 To observe the effects of HH on the proliferation of cultured human hypertrophic scar fibroblasts in vitro. By means of colorimetric MTT(tetrazolium)assay and flow cytometry, we tried to find the effects of HH with different concentration and treating time on the proliferation of cultured human hypertrophic scar fibroblasts. Concentration of HH 0.12g/ml could inhibit proliferation of cultured human hypertrophic scar fibroblasts, Its ID50 is 0.66g/ml. HH could completely inhibit the proliferation of fibroblasts with a concentration of 10g/ml, whereas no effect were found on cultured normal human dermal fibroblasts. HH inhibits proliferation of cultured human hypertrophic scar fibroblasts depending on its concentratione and treating time.
