人白细胞干扰素对子宫内膜腺癌细胞系的作用

摘要 目的观察人白细胞干扰素(IFN-α)对子宫内膜腺癌细胞系(JEC)细胞生长和细胞周期的影响。方法用不同浓度:2000U/ml、500U/ml、100U/ml的IFN-α与JEC共同培养8d,观察JEC细胞生长曲线、细胞凝集素受体表达变化,并用流式细胞仪分析癌系细胞周期。结果和结论IFN-α对JEC细胞的增殖有抑制作用,实验浓度越大,抑制作用越明明,但不影响凝集素受体的表达;IFN-α可使JEC细胞G1/G0阻滞,S期积累和G2/M期减少,前者与IFN-α浓度呈正相关,而后者则为负相关。 Objective To obsterve effect of interferon alpha on cell gtowth and cell cycles of human endometrial adenocarcinoma cell line (JEC) established in our laboratory. Metheds Effect of cell growth curve and lectin receptors on the cells were detected after incubaion with different eoncentration at 2000U/ml, 500U/ml 100U/ml, of interferon alpha for 8 days, and alteration of cell cycle were analysed using flow cytometry. Results and Conclusion Interferon alpha can inhibit the proliferation of JEC and the growth of the cells were dose-dependently suppressed. There were no changes in expression of lectin receptors The cells showed G G0 arrest and Saccumulation, but G2/M decrease, it was postive correlation between the former and the concertration of interferon alpha, however, the two latters were negative corrlation.
