扶正益气中药对脾虚大鼠脑内IL-6Rα水平及其基因表达的影响

Regulation of Interleukin-6 Receptor α Level and Its mRNA Expression in Cerebral Tissues of Spleen Deficiency Rats by Traditional Chinese Medicine for Invigorating Qi and Strengthening Body Resistance

目的研究脾虚大鼠脑内白介素-6受体α(interleukin-6 receptorα,IL-6Rα)水平及其mRNA表达的变化,以及扶正益气中药的干预作用。方法将40只SPF级雄性Wistar大鼠分为正常组、模型组、治疗1组(四君子汤治疗)、治疗2组(玉屏风散治疗)。采用耗气破气、饮食失节、劳倦过度结合的方法,复制脾虚动物模型;采用免疫组织化学法测定大鼠脑内IL-6Rα水平,原位杂交法测定大鼠脑内IL-6RαmRNA表达的变化。结果与正常组比较,模型组大鼠海马CA1区和下丘脑腹侧核IL-6Rα水平及IL-6RαmRNA表达水平显著升高(P<0.01);与模型组比较,治疗1组和治疗2组IL-6Rα水平及IL-6RαmRNA表达水平显著降低(P<0.01);治疗1组与治疗2组IL-6Rα水平及IL-6RαmRNA表达水平比较,差异无显著性。结论脾虚可导致IL-6Rα水平及其mRNA表达水平异常,扶正益气类中药四君子汤、玉屏风散可以通过调节IL-6Rα水平及其mRNA表达水平,改善机体防御功能。

Objective To observe the levels of interleukin-6 receptor a （IL-6Rα） and its mRNA expression in cerebral tissues of spleen deficiency rats, and to explore the effects of traditional Chinese medicine （TCM） for invigorating qi and strengthening body resistance. Methods Forty SPF grade, male Wistar rats were divided into normal group, model group, treatment group 1 （treated with Sijunzi Decoction, a famous TCM for invigorating spleen qi）, and treatment group 2 （treated with Yupingfeng Decoction, a famous TCM for invigorating qi to consolidate superficial resistance）. The animal model of spleen deficiency syndrome was induced by composite factors such as damaging vital qi using purgative drugs, improper diets, and over exertion. The level of IL-6Rα in cerebral tissues was assayed using immunohistochemistry method, and the expression level of IL-6Rα mRNA expression was determined using in situ hybridization. Results Compared with those in normal group, the levels of IL-6Rα and IL-6Rα mRNA expression in CAI region of the hippocampus and ventral nucleus of the hypothalamus in model group were significantly increased （P〈0.01）. Compared with those in model group, the levels of IL-6Rα and IL-6Ra mRNA expression in treatment group 1 and treatment group 2 were significantly decreased （P〈0.01）. There were nonsignificant differences in the levels of IL-6Rα and IL-6Rα mRNA expression between treatment group 1 and treatment group 2. Conclusion Spleen deficiency syndrome could lead to abnormality of IL-6Rα level and IL-6Rα mRNA expression level; Sijunzi Decoction and Yupingfeng Decotion, both of which are famous TCM for invigorating qi and strengthening body resistance, could improve the defense function of human body by regulating the levels of IL-6Rα and IL-6Rα mRNA expression.
