压力超负荷后大鼠心肌核仁组成区嗜银蛋白的动态变化及其意义

Significance of dynamic changes of AgNORs in rat heart following pressure overload

摘要 目的探讨用核仁组成区嗜银蛋白(AgNORs)染色反映压力超负荷后早期心肌肥大的可能性。方法作心肌AgNORs染色并作图像分析进行相对定量,分析其与心肌肥大进程的关系。结果随着大鼠血压的升高,左室重量指数于手术5d开始显著增大(P<0.05),并持续升高。超微结构显示术后1d心肌细胞肌浆网逐渐扩张,术后5d尤以核周和肌膜下明显。线粒体立体计量显示,仅线粒体数密度N在术后5d时显著增多(P<0.05),与心肌肥大相关;AgNORs平均颗粒数N、颗粒面积和积分光密度IOD在术后1d开始出现显著变化,即N显著升高,A、IOD显著降低,这种改变持续至术后30d。结论AgNORs染色可反映心肌细胞核活性早期变化,是显示心肌肥大启动的敏感形态学方法。 Objective To determine the possibility of AgNORs in revealing the early response of cardiac hypertrophy due to pressure overload. Methods After AgNORs staining,image analysis were done on progression of cardiac hypertrophy . Results Accompanying the elevation of arterial pressure,the left ventricular mass index increased significantly and continuously (P<0.05). The sarcoplasmic reticulum enlarged gradually at 1 day,and it was obvious about the nucleous and under the membrane.The number of mitochandria elevated markedly at 5 days after operation (P<0.05).The mean number of AgNORs granules (N) increased,area of granules and integral optioal density (IOD) decreased strikingly at 1 day,and it was kept till 30 days after operation. Conclusions AgNORs staining could reveal the early changes of the activity of cardiac nucleous, and it is a sensitive morphological method to demonstrate the initiation of cardiac hypertrophy.
