Osterix在转移性乳腺癌细胞中的表达及对MMP-2、MMP-9和VEGF启动子活性的影响

Expression of Osterix in Metastatic Breast Cancer Cells and Its Effects on MMP-2,MMP-9 and VEGF Promoter Activities

摘要 目的研究Osterix的表达与乳腺癌细胞转移潜能之间的关系以及检测Osterix对MMP-2、MMP-9和VEGF启动子活性的影响。方法利用Western blot检测不同转移潜能的乳腺癌细胞中Osterix的表达。利用荧光素酶报告基因分析的方法检测Osterix对MMP-2、MMP-9和VEGF启动子活性的影响。结果 Osterix在高转移性乳腺癌细胞MDA-MB361和MDA-MB231中高表达,而在非转移性乳腺癌细胞MCF-7中不表达。Osterix能显著上调MMP-2、MMP-9和VEGF启动子的活性。结论 Osterix在转移性乳腺癌细胞中高表达并能显著上调MMP-2、MMP-9和VEGF启动子的活性。Osterix是否介导乳腺癌、前列腺癌等的骨转移需作进一步研究。 Objective To investigate the relationship between Osterix expression and metastatic potential of breast cancer cells as well as to test the effects of Osterix on the promoter activities of MMP-2, MMP-9 and VEGF. Methods Western blot analysis was performed to examine the expression of Osterix in breast cancer cells with various metastatic potential. Luciferase reporter assays were carried out to test the effects of Osterix on MMP-2,MMP-9 and VEGF promoter activities. Results Osterix is highly expressed in metastatic breast cancer cells MDA-MB 361 and MDA-MB 231 and there was no expression of Osterix in poorly metastatic breast cancer cells MCF-7. Furthermore,Osterix was able to significantly enhance MMP-2,MMP-9 and VEGF promoter activities. Conclusions Osterix is highly expressed in metastatic breast cancer cells and it significantly up-regulates the promoter activities of MMP-2,MMP-9 and VEGF. The role of Osterix in bone metastasis of cancers including breast cancer,prostate cancer,etc needs further investigation.
