染料木苷对FFAs诱导的HepG2细胞胰岛素抵抗的影响 被引量:5

摘要 目的观察染料木苷对游离脂肪酸(FFAs)诱导的HepG2细胞胰岛素抵抗的影响。方法软脂酸诱导HepG2细胞胰岛素抵抗,给予不同浓度的染料木苷(1、2、4μmol/L)、罗格列酮(1μmol/L)干预,采用MTT法检测药物对细胞增殖的影响,以葡萄糖氧化酶法检测葡萄糖消耗量(GC),使用Western blot法检测肝细胞葡萄糖转运体1(GLUT1)蛋白的表达水平。结果不同浓度的染料木苷对HepG2细胞增殖无影响;与对照组比较,软脂酸作用24h后,HepG2细胞在胰岛素刺激下的GC降低,GLUT1表达减少(P<0.05,P<0.01);与模型组比较,染料木苷组HepG2细胞GC随浓度增加而明显提高,GLUT1蛋白表达明显增加(P<0.05,P<0.01)。结论染料木苷能改善软脂酸诱导的HepG2细胞胰岛素抵抗。 Objective To investigate the effects of genistin on improving free fatty acid-induced insulin resistance in HepG2 hepatocytes.Methods Insulin resistance in HepG2 cells was established by adding palmitic acid to the culture medium.Intervention of genistin at different concentration or rosiglitazone was performed at the same time.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT)assay was used to determine the effect of the drugs on cell proliferation.Glucose consumption(GC)was determined by glucose oxidase method.The expression of glucose transporter1(GLUT1)protein was detected by Western blot.Results Genistin at different concentration did not affect cell proliferation.Compared with control group,insulin-stimulated glucose transport in HepG2 cells was inhibited as well as the GC reduced after the intervention of palmitic acid for 24 hours.Meanwhile,GLUT1 expression was obviously reduced(P〈0.05,P〈0.01).However,the above changes were reversed by genistin in a dose-dependent manner(P〈0.05,P〈0.01).Conclusion Free fatty acid-induced insulin resistance in HepG2 hepatocytes might be improved by genistin.
