摘要
目的观察脑外伤后小鼠海马区notch信号基因的表达变化。方法采用Feeney自由落体法复制闭合性脑外伤小鼠模型,分为假手术对照组和脑外伤组,于伤后4h、1d、3d分别处死。采用实时荧光定量PCR法检测伤后各时间点notch1、hes1mRNA的表达。结果 (1)notch1 mRNA表达水平在伤后4h开始升高,1d时达到高峰,较对照组升高约6倍,3d时有所回落,但仍维持在高水平,较对照组升高2倍。(2)hes1 mRNA在伤后4h无明显变化,1d时达到高峰,较对照组升高3倍,3d时回落到正常水平。结论脑外伤后notch1和hes1 mRNA明显增高,提示notch信号被激活,可能与伤后NSCs增殖分化相关。
Objective To explore the expression change of notch signal pathway in hippocampus after traumatic brain injury.Methods Mice were suffered closed head injury by improved device of Feeney’s weight-dropping model and were sacrificed at 4h,1d,and 3d following injury.The mRNA expression levels of notch1 and hes1 at different time points post injury were evaluated by real-time PCR.Results(1) The mRNA expres-sion of notch1 increased at 4h post injury,peaked at 1d post injury,about 6-fold up-regulated change compared with control group,and decreased to 2-fold up-regulated at 3d post injury.(2) The mRNA expression of hes1 didn’t increase at 4h post injury,but increased at 1d post injury,3-fold up-regulated change compared with control group,and decreased to basal levels at 3d post injury.Conclusion The increased mRNA expression lev-els of notch1 and hes1 post injury demonstrate that notch signal pathway may be associated with the process of proliferation and differentiation of NSCs post injury.
