摘要
本文对钝顶螺旋藻藻胆蛋白(包括藻蓝蛋白,缩写C-PC和别藻蓝蛋白,APC)的分离工艺进行了研究,建立经细胞破碎、硫酸铵沉淀蛋白,羟基磷灰石柱分离工艺。经凝胶电泳、吸收光谱、荧光光谱和量热分析等研究,证明 C-PC由大小两个亚基组成,分子量分别为 17 200u和 14 900u;APC由一个亚基组成,分子量为13 400u。其最大吸收峰分别在 625um和 660um处。其荧光发射峰位于 648um。通过 DSC分析发现,APC的变性温度在 49.0℃~60.5℃范围。应用黄嘌呤氧化酶-黄嘌呤-Luminol发光体系证实 APC具有对 O_2-的清除作用。
Two types of phycobiliprotein,C-PC and APC were separated just after the smashed cells were precopitated in (NH_4)_2SO_4 and then loaded on the hydroxylapatite column. The phycobiliprotein' s properties were investigted by methods of electrophoresis, absorption spectrum, fluorescence spectrum and differential scanning calorimetry. The results show that the molecular weights of subunits of the C-PC are 17 200u and 14 900u,APC are 13 400u, Absorption maximum at 625um and 660um,and a room fluorescence emission peak at 648um and 668um. The temperature of denaturation of the APC is in the range of 49. 0 to 60. 5℃. The APC for scaverging active oxygen radical was reported in this paper. The results showed that it could scavenge O_2^-.
