胃癌患者外周血CD4^+ CD25^(high)调节性T细胞比率及表型特征研究 被引量:4

摘要 目的:探讨胃癌患者外周血高表达CD25的CD4+调节性T细胞(CD4+CD25highT)比率变化特点及其临床意义。方法:用流式细胞术检测50例肿瘤患者和20名健康人外周血CD4+CD25highT细胞水平,初步探讨其表型特征。并分析10例胃癌患者手术后1周外周血调节性T细胞与手术前相比的变化。结果:胃癌患者外周血中CD4+CD25highT细胞占CD4+T细胞的百分比率为(14.67±6.55)%,明显高于健康体检者(3.41±1.13)%(P<0.01);CD4+CD25highT细胞具有调节性T细胞的表面标记特征,高水平表达FOXP3、CD152(CTLA-4)、CD134(OX40)和GITR。胃癌各期CD4+CD25highT细胞占CD4+T细胞的比率均显著高于正常对照组(P<0.01)。胃癌患者术后外周血CD4+CD25highT细胞比率与术前相比明显降低(P<0.05)。结论:胃癌患者外周血CD4+CD25highT细胞水平明显升高,可能与肿瘤免疫功能低下及肿瘤发生发展密切相关。 Objective:To study the proportion of CD4+ CD25high regulatory T cells(Treg) in the peripheral blood of patients with gastric cancer and its clinical significance.Methods:The proportion and the phenotype of Treg cells in the peripheral blood of 50 gastric cancer patients and 20 healthy controls were determined by flow cytometry.In addition,the changes of Treg cell levels of 10 patients with gastric cancer were analyzed before and one week after the operation.Results:Compared with the healthy controls(3.41±1.13)%,the proportion of Treg cells in the peripheral blood of patients with gastric cancer was significantly increased(14.67±6.55)%(P0.01).In addition,the CD4+ CD25high Treg cells expressed high levels of FOXP3,CD152(CTLA-4),CD134(OX40) and GITR.The Treg cell level in patients with gastric cancer was significantly reduced after the tumor was resection(P0.05).Conclusions:The proportion of Treg cells is increased in the peripheral blood of patients with gastric cancer;The Treg cells may suppress the immune response against cancer and be related to the progress of the tumor.
