大鼠皮层神经元NMDA受体通道动力学特性的发育变化

摘要 目的:探讨大鼠发育过程中皮层神经元NMDA受体通道动力学特性的变化。方法:采用细胞贴附式记录急性分离神经元NMDA受体的单通道电流。结果:新生大鼠皮层神经元NMDA受体通道以20ps通道开放最常见,而且多数是以短开放为主,猝发样开放(burst)为主的长开放通道少见;成年大鼠20pS通道明显减少,35pS通道明显增多,并且出现35pS长开放通道,20、35pS通道的开放时间和开放概率均明显高于新生大鼠。结论:大鼠皮层神经元NMDA受体通道动力学特性有明显的发育变化。 Objective: To observe the developmental changes of the kinetic properties of NMDA receptor channel in the cortical neurons in rats. Methods: Cell-attached method was employed to record the NMDA receptor channel in the cortical neurons. Results: The 2O pS short open NMDA receptor channel existed in great majority in the cortical neurons of neonatal rats. The number of 2O pS channels was greatly decreased and that of 35 pS ones markedly increased in adult rats in which there were 35 pS long open channels. The open time and open probability were longer and greater in adult rats than in neonatal ones.Oonclusion: There are significantly developmental changes of the kinetic properties of NMDA receptor channels in the cotical neurons in rats.
