纤维蛋白对肾小球内皮细胞表达胞间粘附分子-1的影响及其机制探讨

EFFECTS OF FIBRIN ON ICAM1 EXPRESSION IN GLOMERULAR ENDOTHELIAL CELLS

观察体外培养的肾小球内皮细胞（ＧＥＣ）表面原位形成纤维蛋白凝胶对ＧＥＣ表达胞间粘附分子－１（ＩＣＡＭ－１）的影响。结果：８５％～９３％在无血清ＲＰＭＩ１６４０培养液中培养的ＧＥＣ表达ＩＣＡＭ－１阳性。纤维蛋白显著促进ＧＥＣ表达ＩＣＡＭ－１（Ｐ＜０．０１），呈现明显的剂量、时间依赖关系。放线菌酮和放线菌素Ｄ均可阻断纤维蛋白诱导的ＩＣＡＭ－１表达增强，Ｎｏｒｔｈｅｒｎ杂交显示，纤维蛋白刺激ＧＥＣ４ｈ可诱导ＩＣＡＭ－１ｍＲＮＡ的表达增强。立止血催化形成的ｄｅｓ－ＡＡ－纤维蛋白与凝血酶催化形成的ｄｅｓ－ＡＡＢＢ－纤维蛋白对ＧＥＣ表达ＩＣＡＭ－１的影响差异无显著性意义，而且肝素对纤维蛋白介导的ＩＣＡＭ－１表达增强没有阻抑作用。说明纤维蛋白促进ＧＥＣ表达Ｉ－ＣＡＭ－１呈时间－剂量依赖性，并与基因的转录和翻译有关。

Effects of covering fibrin on the expression of intercellular adhesion molecule1 (ICAM1) in glomerular endothelial cells (GECs) were investigated. Results: 85%￣93% GECs cultured in serumfree RPMI 1640 medium expressed ICAM1 positively. Fibrin gels in situ polymerized with 0.05NIH U/ml thrombin promoted ICAM1 expression on GECs significantly (P<0.01) in a dose and time dependent manner. Both cycloheximide and actinomycin D prevented the upregulation of ICAM1 expression induced by fibrin. Northern blot hybridization showed that fibrin promoted ICAM1 mRNA expression in GECs at 4 hours. DesAA fibrin catalyzed by reptilase and desAABB fibrin catalyzed by thrombin promoted ICAM1 expression insignificantly and heparin had no effect on ICAM1 expression induced by fibrin. It is concluded that fibrin promotes ICAM1 expression in GECs in a dose and time dependent manner, which is both gene transcription and gene translation dependent, but is unlikely related to the heparinbinding domain on fibrin molecules.
