摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞中MBD4 mRNA表达及其与疾病发病机制、疾病活动性的关系。方法以β-actin为内参照,建立逆转录实时荧光定量聚合酶链反应(FQ-RT-PCR)方法,在Light CyclerReal time PCR检测仪上检测70例SLE患者(活动期32例、缓解期38例)、其他结缔组织病(CTD)患者30例(疾病对照组)、健康体检者30例(正常对照组)外周血单个核细胞(PBMCs)中MBD4 mRNA的表达水平和含量。以ΔCt=Ct(待测基因)-Ct(内参基因)来比较基因表达水平的高低。结果 SLE患者PBMC中MBD4 mRNA表达水平高于正常对照组与疾病对照组(P<0.05),且疾病不同病期(活动期与缓解期)MBD4 mRNA表达水平差别有统计学意义(P<0.01)。结论结果显示SLE患者PBMC中MBD4 mRNA表达水平显著高于正常人、疾病对照组,提示MBD4的表达与SLE的发生发展存在一定关联性,可能参与了SLE的发病过程并与疾病的活动性有关。
Objective The objection of this study is to explore the expression of MBD4 mRNA in peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus, and its polymerase chain reaction (FQ-RT-PCR) was used to examine MBD4 mRNA expression on peripheral blood monocytes of 70 cases from SLE(32 cases in active stage,38 cases in stable stage),27 cases of other CTD as diseases control, and 27 cases as normal controls, with β-actin as an endogenous control. The levels of the mRNA expression were calculated with the formular ACt = Ct(objiective gene) - Ct(pastin). The mean MBD4 mRNA expression of SLE in active stage was higher than that of other CTD and healthy controls( P 〈 0.05). There was statistical difference between the expressions in active and inactive SLE patients those ( P 〈 0.01). Results MBD4 mRNA expression levels on PBMCs is significantly elevated in patients with SLE, and it maybe correlated to pathogenesis and activity disease.
