新疆天山雪莲根际冻土微生物富集培养的初步研究 被引量:5

摘要 利用纤维素、豆粕、魔芋粉、木聚糖等为底物对新疆天山雪莲根际冻土微生物进行了低温富集培养与分离。通过16S rDNA序列扩增初步鉴定了分离所得微生物,并构建系统发育树。结合选择性培养基进行了产纤维素酶、α-半乳糖苷酶、β-甘露聚糖酶和β-木聚糖酶等微生物的筛选。结果表明,新疆天山雪莲根际冻土中蕴含着丰富的产酶微生物,其中以假单孢菌属、黄杆菌属、根瘤菌属和不动杆菌属等属的微生物为优势菌群。分离得到的41株细菌中大部分都能产水解酶类。 The microflora in the frozen soil at the root of Saussureae Involucratae Kar.et Kir.et Maxim from Tianshan in Xinjiang Autonomous Region were isolated by enrichment culture using cellulose,soybean,konjac powder and xylan as substrate,and identified by 16S rDNA sequences amplification,the neighbor-joining phylogenetic tree was then constructed.The microflora producing all kinds of enzymes including Cellulase,α-galactosidase,β-mannanase,β-xylanase ete,was screened by selective mesa.The results showed that rich enzyme-producing microflora existed in the frozen soil,while Pseudomonas,Flavobacterium,Rhizobium and Pedobacter are the advantage bacterium groups.Among 41 bacteria strains isolated,most of them excreted hydrolysis enzyme.
