摘要
利用偏磷酸溶液提取,以0.01 moL/L磷酸二氢钾溶液(pH值2.5-2.8)为流动相,亲水性Venusil MP C18色谱柱分离,二极管阵列检测器测定,检测波长245 nm,建立准确快速的高效液相色谱仪检测肉制品中L-抗坏血酸与D-异抗坏血酸含量的方法。结果表明,L-抗坏血酸与D-异抗坏血酸可以达到基线分离,在2-100 mg/L范围内线性相关系数均〉0.999;10-100 mg/kg浓度水平添加回收率为79.3-105.1%,相对标准偏差4.5%-13.6%(n=6)。此法可以准确快速地分析肉制品中L-抗坏血酸含量与D-异抗坏血酸。
The method was developed with the condition as follows:the sample was extracted with Metaphosphoric acid solution,HPLC analysis was performed on a Venusil MP C18 column with DAD detector,0.01moL/L KH2PO4 solution was used as mobile phase(pH 2.5- 2.8) to establish a quick,accurate method of HPLC to determine L-ascorbic acid and D-isoascorbic acid in meat products.The results showed that the linear ranges were from 2 mg/L to 100 mg/L with correlation coefficients of 0.999.The mean recoveries were from 79.3% to 105.1% at concentration from 10 mg/kg to 100 mg/kg,and the RSDs were from 4.5% to 13.6%(n=6).It is concluded that this method is quick,accurate for determination of L-ascorbic acid and D-isoascorbic acid in meat products.
