大黄素温敏凝胶对人牙周膜细胞的毒性作用 被引量:4

摘要 目的观察大黄素温敏凝胶对人牙周膜细胞的毒性作用,为其治疗牙周病提供依据。方法人牙周膜细胞在不同浓度的大黄素温敏凝胶中体外培养24、48、72、96h,用四甲基偶氮唑盐(MTT)比色法测定细胞相对增殖率(RGR),判断细胞毒性的级别。结果各组细胞不同时间点相对增殖率均在96%~188%之间,各浓度的大黄素温敏凝胶材料浸提液的细胞毒性均为0级或1级,完全符合生物材料的安全评价标准。结论大黄素温敏凝胶对人牙周膜细胞无毒性,用于牙周病局部治疗具有进一步的研究价值。 Objective To evaluate the cytotoxicity of the emodin thermosensitive hydrogel on human periodontal ligament cells (HPDLCs) and provide evidence about local application of the emodin thermosensitive hydrogel on periodontal diseases.Methods HPDLCs were cultured in medium containing the emodin thermosensitive hydrogel at different concentrations for 24 h,48 h,72 h,96 h and the relative growth rates (RGR) were determined by MTT assay for evaluation of cytotoxicity.Results The RGF of the HPDLCs are 96%~188% at 24 h,48 h,72 h,96 h and cytotoxicity grade of different concentrations of the emodin thermosensitive hydrogel is 0 and 1.Conclusion The RGR showed no cytotoxicity on HPDLCs.Local application of the emodin thermosensitive hydrogel for treating periodontal diseases is feasible.
