摘要
目的:目的:观察针刀刺激对幼鼠缺血缺氧性脑损伤的影响。方法:新生7日龄SD幼鼠28只,随机分为手术组20只及假手术组8只。手术组幼鼠复制为缺血缺氧性脑损伤模型,共成功16只,再随机人为手术组A和手术组B各8只。手术组A于造模后2、9及16d时参照《实验针灸学》取百会、大椎、曲池、内关、环跳、阳陵泉及三阴交穴分别交替给予针刀治疗1次。造模第17天时通过悬吊试验观察各组随意运动功能,免疫组化观察脑组织缝隙连接蛋白(CX32)及巢蛋白(Nestin)的表达。结果:悬吊试验中,手术组A平均悬吊时间评分与假手术组比较差异不明显;手术组B则明显低于假手术组(P<0.05)。CX32表达,手术组B明显低于手术组A及假手术组(P<0.05),手术组A与假手术组比较差异无统计学意义。Nestin表达,手术组A与假手术组及手术组B比较均明显升高(P<0.01)。结论:针刀治疗缺血缺氧性脑损伤可改善随意运动功能障碍,启动内源性神经干细胞的增殖,提高损伤后脑组织CX32的表达。
Objective:To observe the effects of acupotomy therapy on hypoxia-ischemic brain damage (HIBD) in neonatal rats. Methods:Twenty-eight 7-day-old SD rats were randomly divided into 3 groups:sham operation group,HIBD group and acupotomy group. After the models were made successfully,acupotomy therapy was given since the second day,and then once every 7 days. In the 24-day-old rats the voluntary motor function was observed in hanging test,and their brains were removed for detection of gap junction connexin32 (CX32) and nestin expression by using immunohistochemistry. Results:(1) There was significant difference in hanging test between HIBD group and acupotomy group (P0.05),but no difference was found between sham operation group and acupotomy group. (2) The number of CX32 positive cells was decreased dramatically in HIBD group as compared with other two groups (P0.05),and no difference was found between sham operation group and acupotomy group (P0.05). Nestin expression in acupotomy group was increased as compared with sham operation group (P0.001). Conclusion:Acupotomy therapy can be used to improve disability of voluntary motor function,and increase the expression of CX32 and Nestin in brain tissues in HIBD.
