摘要
目的:通过研究叉头框C2(Foxc2)和淋巴管内皮透明质酸受体-1(LYVE-1)在胚胎淋巴管的表达模式,探讨Foxc2在小鼠胚胎淋巴管发育中的作用.方法:以LYVE-1为淋巴管特异标识物,用免疫组织化学方法研究Foxc2在小鼠胚胎淋巴管发生过程的表达模式.结果:从E10.5开始,在小鼠胚胎颈、胸部背主动脉旁出现Foxc2,呈强阳性表达的淋巴囊.E13.5时,在胸段脊椎两侧、脊椎腹侧的软组织和脊椎背侧的体表外胚层等组织中形成小淋巴管网,但无Foxc2染色.小淋巴管沿体纵轴方向延伸,其间有横向毛细淋巴管联系.E15.5以后,Foxc2蛋白仍在脊椎腹侧大的淋巴管内壁内皮细胞核强表达.在胚胎发育后期,表皮下毛细淋巴管数量增多,中、小淋巴管形成,但皮下以及其他部位的毛细淋巴管则未见Foxc2表达.结论: Foxc2参与了小鼠胚胎早期胸、颈部淋巴囊和中心大淋巴管的形成,胚胎中期的大淋巴管塑形仍需要Foxc2.
Objective: To explore the expression pattern of forkhead box C2 (Foxc2) and lymphatic vessel endothelial hyalu- ronan receptor-1 (LYVE- 1) on fetal lymphangion to understand the effect of Foxc2 on lyrnphangiogenesis of mouse fetuses. Methods: LYVE-1 was applied as the special marker of lymphangion and the expression pattern of Foxc2 protein was ana- lyzed with immunohistochemical staining in the lymphangiogenesis of mouse embryo. Results.. On E10.5, saccus lymphaticus stained strongly with Foxc2 antibody were seen on bilateralis of dorsal aorta in the cervical and thoracic region. On E13.5, small lymphatic vessel networks emerged in peripheric soft tissue of vertebrae in thoracic region, but Foxc2 stain was negative. The small lymphatic vessels extended along body longitudinal axis, which was connected with transverse lymphatic capillaries. After El5.5, Foxc2 protein still strongly expressed in endothelial cell nuclei of lymphangion lying in the ventral side of vertebrae. During the later stage of fetal development, the number of lymphatic capillaries increased and the medium and small lymphatic vessels formed in subepidermal tissue; but Foxc2 expression was not found in the lymphatic capillaries of subepidermal tissue or in the lymphatic capillaries of other tissues. Conclusion: The Foxc2 protein is involved in the formation of saecus lymphaticus and large central lymphatic vessels in the cervical and thoracic region of mouse during the early stage of embryo development. The large lymphatic vessel remodeling still requires Foxc2 during the middle stage fetal development.
