摘要
目的探讨依达拉奉(Edaravone,Eda)对谷氨酸(Glutamate,Glu)诱导皮层神经元释放一氧化氮的影响。方法 SD乳鼠皮层神经元原代培养7d后,分对照组、Glu组(50μM)以及Glu(50μM)+Eda(100μM)组,分别在加药前、加药6h和24h后取三组培养基以硝酸还原酶法检测NO浓度。结果加药前三组培养基的NO浓度没有显著差异(P>0.05),加药6h、24h后,Glu组和Glu+Eda组NO浓度均显著高于对照组(P<0.01),而且Glu组NO浓度显著高于Glu+Eda组(P<0.01)。结论依达拉奉能降低谷氨酸诱导皮层神经元释放一氧化氮。
Objective To explore the effect of edaravone (Eda) on nitric oxide (NO) release in cultured cortical neurons stimulated by glutamate (Glu).Methods Primary cotical neuron cultures were prepared from neonate SD rat brain and incubated for 7 days into confluence. The cultured cortical neurons were divided into three groups,control group,Glu (50 μM) group and Glu (50 μM)+ Eda (100 μM) group. The medium was collected at 0 h,6 h,24 h interval for detection of NO concentration using nitrate reductase method.Results No significant difference of NO concentration was observed among each groups at 0 h. NO concentration significantly increased in Glu group and Glu + Eda group at both 6 and 24 h than that in control group (P〈0.01). And the NO concentration in Glu group was even higher than that in Glu + Eda group at both 6 and 24 h (P〈0.01).Conclusion Edaravone can attenuate the NO release of cultured cortical neurons stimulated by glutamate.
