SCH66336对胃癌细胞增殖和凋亡的影响及作用机制 被引量:3

摘要 目的探讨法尼基转移酶抑制剂(FTIs)SCH66336对胃癌SGC-7901与MGC-803细胞增殖、凋亡的影响及机制。方法取对数生长期胃癌SGC-7901与MGC-803细胞,分为实验组和对照组,实验组加入SCH66336使终浓度分别为0.1、0.3、1.0、3.0μM,对照组加入含等体积DMSO的培养基,每组设3个复孔。MTT法检测各组细胞增殖抑制率;流式细胞术测定细胞周期与细胞凋亡率,Western blot法检测细胞中核糖体蛋白S6的磷酸化(S235/236)、细胞周期素D1(cyclin D1)与活化的Caspase-3表达情况。结果与对照组比较,SCH66336作用后SGC-7901与MGC-803细胞抑制率及凋亡率明显升高,呈G0/G1细胞周期阻滞,cyclin D1与S6磷酸化表达明显降低、活化的Caspase-3表达升高(P<0.05),且呈时间及剂量依赖性。结论 SCH66336对SGC-7901与MGC-803胃癌细胞具有增殖抑制和凋亡诱导作用,其机制可能为抑制S6的活化、下调cyclin D1表达、激活Caspase-3依赖的凋亡通路。
