摘要
建立了以无醛乙醇为溶剂,采用碱、酸两步水解和紫外分光光度法测定大豆磷脂中总糖含量的方法.结果表明:大豆浓缩磷脂中总糖含量为3.48%,粉末大豆磷脂中总糖含量为3.22%,乙醇法提取的磷脂中总糖含量为3.52%,乙醇冷冻法纯化分离的磷脂中总糖含量为3.96%,经柱层析纯化分离的磷脂酰胆碱(w(PC)=90%)试样中总糖含量未检出;并结合薄层色谱法进一步分析测定了前4种磷脂试样中糖脂组分及含量,糖脂主要为单半乳糖甘二酯和单半乳糖甘一酯,总糖脂含量分别为4.29%、3.75%、4.44%、5.41%.
We put forward a method for determining total carbohydrate content in soybean phospholipids by acid hydrolysis, alkali hydrolysis and ultraviolet spectrophotometry, selecting aldehyde-free ethanol as solvent. The results show that the total carbohydrate content in concentrated soybean phospholipids is 3.52% ; the total carbohydrate content in powdered soybean phospholipids is 3.22% ; the total carbohydrate content in soybean phospholipids obtained by ethanol extraction is 3.52% ; the total carbohydrate content in soybean phospholipids purified and separated by chilled ethanol extraction method is 3.96% ; and no total carbohydrate is detected in the phosphatidylcholine (w(PC) = 90% ) separated and purified by column chromatography. Furthermore, we also determined the composition and content of glycolipids in the above four soybean phospholipids samples by thin-layer chromatography; and the results show that the glycolipids mainly contain monogalactosyl diglyceride and monogalactosyl monoglyceride, and the contents of the total glycolipids are respectively 4. 29%, 3.75%, 4.44% and 5.41%.
