摘要
目的建立高效液相色谱法(high-performance liquid chromatography,HPLC)测定万艾可片中枸橼酸西地那非含量的方法。方法 C18色谱柱(4.6mm×250mm,5μm),以水-甲醇(2∶8)为流动相;流速为1ml/min,检测波长为290nm。结果回归方程为Y=1197.454 X+14.984,r=0.9999,线性范围1.1392~5.6960μg,平均加样回收率为100.08%(RSD=1.36%),样品在12h内稳定。结论本方法结果准确可靠,可用于万艾可片的质量控制。
Objective To establish a method for determining sildenafil citrate contents in Viagra by high-performance liquid chromatography (HPLC). Methods The chromatographic conditions were: diamonsil C18 column (4.6 mm×250 mm, 5μm), methanol-water (8∶2) as the mobile phase at a flow rate of 1ml/min and a detective wavelength at 290 nm. Results Sildenafil citrate showed a good linear relationship at the range of 1.1392~5.6960μg, the linear equation was Y=1197.454X+14.984 (r=0.9999), the recovery rate was 100.08% (RSD=1.36%) and the sample was stable within 12 hours.Conclusion The method is sensitive, accurate, reproducible, specific and can be efficiently used for quality control of sildenafil citrate in Viagra.
