摘要
目的研究腺病毒介导的PDGF-BB的基因治疗对大鼠大脑中动脉闭塞(MCAO)模型的治疗作用及脑血管发生的影响。方法采用改良的Zea Longa法制备大鼠脑缺血模型,将30只SD大鼠随机分为假手术组,对照组(1 acZ组)和治疗组(Ad-PDGF-BB组),于造模成功后24h尾静脉给药。检测实验各组神经功能评分恢复情况及脑组织含水率的变化,通过核磁共振和免疫组化分别检测实验各组大脑梗死体积和血管发生的变化。结果治疗组大鼠神经功能恢复对比对照组有显著改善,脑组织含水率和脑梗死面积明显小于对照组,脑血管密度显著高于对照组。结论腺病毒介导的PDGF-BB基因能够显著减小脑组织含水率和脑梗死体积,促进脑部血管发生,为缺血性脑卒中的基因治疗提供了新的途径。
Objective To study the effect of Adenovirus mediated PDGF-BB gene on the middle cerebral artery occlusive rats model and treated in acute ischemic cerebrovascular disease.Methods Thirty rats were randomly divided into three groups,i.e.,sham group,control group(lacZ group) and drug treatment group(Ad-PDGF-BB group).The model of middle cerebral artery occlusion in rats was established successfully by Zea Longa method.Twenty-four hours later,Ad-PDGF-BB was given to the rats in Ad-PDGF-BB group through intravenous injection.The Ad-lacZ was given to the rats of the control group.The neurological deficit scoring,the water content,the rats' ischemic volume and the change of microvessel density in the brain of middle cerebral artery occlusion rats were detected.Results In the treated group,the recovery of neural function was improved significantly,the water ratio and infarct size of brain were decreased obviously and the microvessel density increased remarkably,comparing with the control group,there were significant improvement in the neurological deficit scoring,the water ratio and the rats′ ischemic volume of the drug treatment group and the microvessel density increased remarkably.Conclusion The Adenovirus mediated PDGF-BB gene has obvious effect on the decreasing the water ratio and the rats′ ischemic volume,and promoting angiogenesis.These findings may provide a new strategy in treating acute ischemic cerebrovascular disease with gene therapy.
